The X chromosome is a unique genetic environment. Female cells contain two X chromosomes while male cells contain only one, and to balance the X-gene dose in female cells one X copy is kept transcriptionally silenced by the process of X-chromosome inactivation. This is achieved by remarkable remodeling of the inactive X chromosome, which drastically changes its chromatin structure, methylation patterns, and three-dimensional conformation – ultimately suppressing the expression of most of its genes. By characterizing the epigenetic framework of X-chromosome inactivation, we reveal fundamental features of gene regulation. During the last years Reinius and colleagues pioneered the use of single-cell RNA-seq in the study of X-inactivation (Science 2014, Cell 2016, Nature Genetics 2016 among other publications). Currently the Reinius lab is working to determine the X-inactive landscape across a wide range of in vivo tissues and cell types. This is accomplished by a comprehensive synthesis of newly developed concepts and techniques, including single-cell RNA-sequencing, chromatin accessibility and modification assays, mouse genetics and transgenics, and recently advanced computational methods for the analysis of allele-specific gene expression. The expected outcome of the research is of broad biological significance, and may reveal gene-regulatory features underlying variable expressivity and incomplete penetrance of X-linked traits and genetic disorders.

A postdoctoral position is available in the Reinius lab. The applicant is a highly motivated, creative and interactive person with a passion for understanding biology in depth. The main research focus will be to characterize transcriptional and epigenetic states of X-chromosome inactivation. Key methodologies include PCR-based amplification techniques, RNA/DNA-sequencing, molecular cloning, immunoprecipitation assays, fluorescence and confocal microscopy

The Department of Medical Biochemistry and Biophysics conducts multidisciplinary research in biomedicine and education within biochemistry and biophysics. The Reinius laboratory is placed in the Division of Biomaterials; an international, interactive, young and vibrant research environment. This Division is located at Biomedicum, KI Solna Campus, a modern, purpose-built facility for experimental medical research. It hosts research groups with diverse backgrounds, forming a unique cluster with expertise in cellular and molecular biology, high-content RNA/DNA sequencing, epigenetics, bioinformatics, high-resolution imaging and novel DNA-based detection and imaging techniques, and many other subjects. Excellent research facilities are available.

The postdoctoral researcher will perform the work as part of the Reinius group at the Department of Medical Biochemistry and Biophysics. The main assignment is experimental work (molecular biology and biochemistry). Some administrative duties will also be assigned

Entry requirements
Qualified to be employed as a postdoctor is one who has obtained a doctorate or has equivalent scientific competence. In assessing candidates we will place particular emphasis on the personal trajectory and skills. To qualify, the applicant must hold a recent PhD degree and have a strong motivation to pursue challenging research. The ideal candidate should have demonstrated skills in PCR-based methodology, microscopy, RNA/DNA-FISH, and molecular cloning. Experience in methods development and/or biochemistry is a plus. The optimal candidate will be open to new ideas, flexible, engaged and goal oriented. The applicant should have good communication skills and be proficient in spoken and written English.
