The Department of Cell and Molecular Biology (CMB) is a department with a strong focus on basic science. CMB conducts research and education in cell biology, molecular biology, developmental biology, stem cell biology, cancer and infection biology.

An exciting postdoctoral project is available in the group of Laura Baranello at the Department of Cell and Molecular Biology (CMB), Solna. The group focuses on understanding how transcription and DNA topology are functionally coordinated in human cells, and uses a combination of approaches spanning from biochemistry, molecular biology, next-generation sequencing and bioinformatics. The group is also associated to the laboratory of David Levens at the National Institutes of Health in US. The successful applicant will thus be part of multiple networks of recognized scientists, and have access to state-of-the-art infrastructure and facilities.

Topoisomerases are essential enzymes that regulate the topology of DNA favoring DNA replication and RNA transcription  by cleaving and resealing DNA strands   (Baranello et al., Transcription, 2013; Kouzine et al., Nat. Struct. Mol. Biol., 2013). Because cancer cells have higher rates of replication and global transcription compared to normal cells, they are highly dependent on topoisomerase activity. Therefore, strategies to target topoisomerases could provide a mean to selectively affect cancer cells.

This interdisciplinary research project aims to define the role for topoisomerases in transcription, to understand how topoisomerases are regulated and to search for drug-like candidates that target these regulatory mechanisms. Recent evidence show that topoisomerases execute their function through regulatory interactions with partner proteins that modulate their activity to affect the transcriptional outcome (Baranello et al., Cell, 2016). We will use biochemical assays, DNA and RNA sequencing approaches, CRISPR-based genome editing as well as drug screens, to reveal new factors regulating topoisomerase activity, decipher the molecular mechanisms of topoisomerase regulation and identify drugs targeting their interactions with topoisomerases in cancer cells.

Entry requirements
Scholarships for carrying out postdoctoral research may be granted to persons living in other countries who come to Sweden with the intention of only staying in Sweden for all or part of their postdoctoral education at KI. Persons who are resident in Sweden and/or have carried out all or part of their academic education at KI or some other educational institution in Sweden cannot be considered for scholarships from KI. The head of the department determines whether their previous training and scholarly qualifications correspond to a Swedish PhD or higher.

We are looking for a curious, resourceful and highly motivated candidate with a PhD in molecular biology or related sciences. The ideal candidate is expected to lead the project in collaboration with the PI and other team members, supervise students and participate in common laboratory duties. We expect the candidate to have a strong scientific background in chromatin biology with relevant expertise in biochemistry and molecular biology techniques and willing to learn and develop sequencing techniques. Experience with large-scale bioinformatic analysis is a strength. Good knowledge of both spoken and written English is a requirement.

Type of scholarship
A scholarship for carrying out postdoctoral research can be granted for a maximum of two years within a four year period following the receipt of a doctoral degree or equivalent. This educational scholarship is tax-exempt.

The amount is set for twelve months at a time and is paid out on a monthly basis. In exceptional cases, shorter periods may be acceptable.
